Tissue Procurement: Human bone tissue is collected from cadaveric donors within 24 hours of death or as surgical specimens from a relevant procedure. The tissue is harvested from the ends of the tibia and femur from both clinically “normal” donors and from those diagnosed with rheumatoid and osteoarthritis. Proper consent and serological information is obtained on all specimens.
Monolayer Culture Processing: Harvested bone tissue is carefully dissected away from surrounding tissues and washed thoroughly to remove blood and debris. An enzyme treatment is applied to isolate primary bone cells. The cells are seeded onto flasks or multi-well plates in Ham’s F12 media supplemented with 10% FBS. The cells are maintained in a humidified incubator at 37oC and 5% CO2 for 48 hours to promote cell adhesion and then shipped with cold packs for overnight delivery. |
